
While semaglutide, tirzepatide and retatrutide dominate the conversation around metabolic peptide research, another compound has already reached the market in China. Here’s what the research is showing about mazdutide.
If you follow developments in GLP-1 research, you have probably heard of semaglutide, tirzepatide and, more recently, retatrutide.
These compounds have become some of the most discussed names in modern metabolic research.
But there is another peptide that has received considerably less attention in the UK and much of the Western research community:
Mazdutide.
Mazdutide is particularly interesting because it takes a slightly different approach from semaglutide and tirzepatide.
It activates both the GLP-1 receptor and the glucagon receptor, making it a dual-receptor agonist.
And unlike many of the newer compounds still being investigated, mazdutide has already received regulatory approval in China.
So what exactly is mazdutide, what have the clinical studies shown so far, and why isn’t it talked about more widely in the West?
What Is Mazdutide?
Mazdutide is a once-weekly peptide that activates two important metabolic signalling pathways:
- GLP-1 receptor
- Glucagon receptor
This makes its mechanism different from some of the better-known compounds.
Semaglutide primarily targets the GLP-1 receptor, while tirzepatide targets both the GLP-1 and GIP receptors.
Mazdutide instead combines GLP-1 and glucagon receptor activity.
This is important because glucagon has biological effects that extend beyond appetite and glucose regulation. It is involved in processes including energy expenditure and lipid metabolism, which has made the combination of GLP-1 and glucagon receptor activity an interesting area of research.
In simple terms, researchers have been investigating whether combining these pathways can produce a broader metabolic effect than targeting GLP-1 alone.
Mazdutide Is Already Approved in China
This is probably the most interesting part of the story.
Mazdutide received its first approval from China’s National Medical Products Administration in June 2025 for long-term weight management in adults with overweight or obesity.
It subsequently received approval in China in September 2025 for glycaemic control in adults with type 2 diabetes.
That makes mazdutide quite different from compounds such as retatrutide, which remains investigational.
It also means that mazdutide has moved beyond the purely experimental stage in at least one major pharmaceutical market.
The approval was supported by clinical research conducted in Chinese adults, including the large Phase 3 GLORY-1 study.
What Did the GLORY-1 Study Show?
One of the most important studies of mazdutide so far is the GLORY-1 Phase 3 trial, published in the New England Journal of Medicine in 2025.
The study included 610 adults with overweight or obesity in China.
Participants were randomly assigned to receive:
- 4 mg mazdutide
- 6 mg mazdutide
- placebo
The study continued for 48 weeks.
The results were notable.

The researchers also reported that 35.7% of participants receiving 4 mg and 49.5% receiving 6 mg achieved at least a 15% reduction in body weight by week 48.
The study also reported improvements across the prespecified cardiometabolic measurements.
As with other GLP-1-related compounds, gastrointestinal adverse events were among the most frequently reported, and were generally mild to moderate in the study.
Importantly, these results came from a controlled clinical trial population. They shouldn’t be interpreted as predicting what an individual person would experience.
What About Diabetes Research?
Mazdutide has also been studied specifically in adults with type 2 diabetes.
A Phase 3 study published in Nature included 320 Chinese adults with type 2 diabetes who were inadequately controlled with diet and exercise.
After 24 weeks, HbA1c fell by:
- 1.57 percentage points with 4 mg
- 2.15 percentage points with 6 mg
- compared with 0.14 percentage points with placebo.
Body weight also decreased by approximately:
- 5.61% with 4 mg
- 7.81% with 6 mg
- compared with 1.26% with placebo
This provides another interesting piece of evidence that mazdutide’s effects aren’t limited to body-weight research.
The combination of glucose-related and weight-related outcomes is one reason the compound has attracted attention within metabolic research.
Mazdutide Has Now Been Studied Outside China Too
This is where the story becomes particularly interesting for Western researchers.
Mazdutide is sometimes described as a mainly Chinese development, but that is becoming less accurate.
A U.S.-based Phase 2 trial was published in The Lancet Diabetes & Endocrinology.
The study involved 179 adults with overweight or obesity without type 2 diabetes across 24 U.S. centres.
Researchers examined mazdutide across several dose ranges over 48 weeks.
At week 32, average body-weight changes were:

Additional weight reduction was observed by week 48.
The researchers reported that gastrointestinal events were the most common adverse events. Treatment discontinuation because of adverse events occurred most frequently in the 16 mg group, where it was reported in 20% of participants, primarily because of gastrointestinal disorders.
This U.S. study is particularly relevant because it provides clinical data from outside the original Chinese development programme.
So Where Does Mazdutide Fit Alongside the Other Peptides?
This is probably the easiest way to understand why researchers are interested in it.

The interesting comparison is between mazdutide and retatrutide.
Both involve glucagon receptor activity.
However, retatrutide adds GIP receptor activity as well, giving it a three-receptor mechanism.
That doesn’t automatically mean that one approach is better than the other.
It simply means researchers are investigating different combinations of metabolic signalling.
And this is where the field becomes particularly interesting.
Why Does Glucagon Receptor Activity Matter?
GLP-1 has become extremely well known because of its role in appetite regulation, glucose control and other metabolic processes.
Glucagon is different.
It has an important role in regulating energy availability and metabolism, including effects involving the liver and energy expenditure.
The theory behind combining GLP-1 and glucagon receptor activity is therefore different from simply increasing GLP-1 activity.
Researchers are essentially asking:
Can activating both pathways produce a broader metabolic response than activating GLP-1 alone?
The clinical studies so far suggest that this approach can produce substantial changes in body weight and metabolic measurements, although the precise contribution of each pathway is still an important area of research.
That distinction is important.
A peptide having an interesting mechanism doesn’t automatically tell us exactly how every biological effect is produced.
Why Haven’t We Heard More About Mazdutide in the West?
This is one of the more interesting questions.
There are several factors worth considering.
First, the Western GLP-1 conversation has been dominated by the enormous clinical and commercial attention surrounding semaglutide and tirzepatide.
More recently, attention has shifted towards investigational compounds such as retatrutide.
Mazdutide has therefore received considerably less attention in mainstream Western discussions.
There is also a geographical element.
Its first approval was in China, and much of the early Phase 3 evidence came from Chinese clinical populations.
That doesn’t make the research less interesting.
In fact, the publication of clinical research involving participants in the United States suggests that mazdutide is now receiving broader international attention.
It may simply be that the Western research community is only beginning to catch up with a compound that has already progressed considerably in China.
Mazdutide vs the “Big Three”
When people talk about the major metabolic peptides today, the conversation usually centres around:
Semaglutide → GLP-1
Tirzepatide → GLP-1 + GIP
Retatrutide → GLP-1 + GIP + glucagon
Mazdutide adds another interesting position:
Mazdutide → GLP-1 + glucagon
That makes it particularly relevant when looking at the evolution of multi-receptor peptide research.
The progression is not simply about creating stronger versions of existing compounds.
Researchers are experimenting with different combinations of receptor activity to understand how those combinations influence metabolism.
Mazdutide is an excellent example of that approach.
What We Still Don’t Know
Despite the encouraging clinical data, there are still plenty of unanswered questions.
Longer-term research will help clarify how sustained the effects are over several years and how different metabolic outcomes develop over time.
There is also considerable interest in potential applications beyond weight management and diabetes.
Mazdutide is being investigated in areas including metabolic dysfunction-associated fatty liver disease, obstructive sleep apnoea and alcohol use disorder, although these remain areas of clinical investigation rather than established indications.
There is also an important distinction between having a compound approved in one country and having it approved elsewhere.
Mazdutide’s approval in China does not mean that it is approved for general clinical use in the UK.
Regulatory status is country-specific.
That is particularly important as interest in newer research peptides continues to grow internationally.
Could Mazdutide Become More Important in Peptide Research?
It certainly makes sense to keep an eye on it.
The combination of:
- GLP-1 receptor activity
- glucagon receptor activity
- Phase 3 clinical data
- approval in China
- additional international clinical research
- ongoing investigation into other metabolic conditions
makes mazdutide an interesting compound to follow.
It also represents something larger happening within peptide research.
The field is moving beyond single-receptor approaches and increasingly exploring multi-receptor signalling.
Tirzepatide demonstrated the potential of combining GLP-1 and GIP activity.
Retatrutide is investigating a three-receptor approach.
Mazdutide provides another route by combining GLP-1 and glucagon signalling.
For researchers watching where the field goes next, that makes it a compound worth knowing about.
What Does This Mean for Research?
For laboratories interested in metabolic signalling, mazdutide provides another useful compound for understanding how different receptor combinations influence biological processes.
Its development also provides an interesting comparison with semaglutide, tirzepatide and retatrutide.
As more international studies are published, we should have a clearer picture of where mazdutide fits within the rapidly developing field of metabolic peptide research.
